The defensive players can take any position they want and can move around on their side of the football before a play starts.

The offensive player has a lot of rules for their positions and what happens on offense. Seven players must line up at the line of scrimmage; the other 4 players must be lined up at least 1 yard behind the line of scrimmage. The offensive players must be still before the play starts with exception to the 4 backs which can move parallel or away from the line of scrimmage.

There are several football penalties, but here are some the important and common ones you might recognize at football game.
The false start is when a football player on the offense moves just before the snap.

Offside is when a player from the offense or defense is on the wrong side of the line of scrimmage at the time of the snap. . A defensive player can cross the line of scrimmage as long as they get back before the snap, but if they touch an offensive player they can be called for encroachment.

Pass Interference is when the defender contacts a pass receiver after the ball is in the air to prevent him from catching the ball. This is up to the referee to determine.

And you have to know a little about the field. The football field is 120 yards long and 53 1/2yards wide. The field is divided up every 5 yards by a yard line. The football is always placed on or between the hash marks at the start of each play.
